Research

Functional analysis of networks of GABAergic interneurons in primary somatosensory (barrel) cortex

GABA-releasing interneurons are substantially involved in information processing in the neocortex and are directly involved in the somatosensory circuitry. Martinotti cells, a readily identifiable type of GABAergic interneuron, are thought to play a central role in modulating dendritic excitation of pyramidal cells because of their morphological structure. Thus, they possess a direct influence on the neocortical output structures of the barrel cortex. In addition to excitatory inputs from surrounding pyramidal cells, Martinotti cells are the target of marked behavioral inhibition, the neocortical origin of which is unclear. In this study, the cells of origin of inhibitory projections to Martinotti cells of neocortical layers 2/3 and 5 in barrel cortex will be investigated using combined whole-cell patch-clamp derivations and local photolysis of caged-glutamate in acute brain slices. Subsequently, "paired" patch-clamp derivations from Martinotti cells and their directly associated GABAergic interneurons will be used to uniquely identify and characterize these interneurons. The knowledge gained will contribute to the model of sensory information processing in the barrel cortex and improve the understanding of the networks of GABAergic interneurons.

Research Objectives:

  • Patch-clamp recordings from Martinotti cells and identify the neocortical layers from which neurons inhibitively project to Martinotti cells using "caged-glutamate" photolysis.
  • Establishment of the method to identify connected GABAergic interneurons in the selected (I) neocortical layers in acute brain slice.
  • Simultaneous "paired" patch-clamp recordings from Martinotti cells and the upstream inhibitory interneurons, and their characterization according to the Petilla terminology.
